چکیده

The polyols erythritol and adonitol reduced 45°heat killing in asynchronous Chinese hamster ovary cells. Heat protection by glycerol and erythritol increased with the apparent intracellular concentration, as inferred from cell volume measurements, and the number of hydroxyl groups per alcohol molecule. The nonlin ear tetrahydroxy alcohol pentaerythritol did not protect but sen sitized to heat killing. On cell survival curves, the reduced cell killing of protected cells was expressed by an increased 00 for the pentahydroxy alcohol adonitol (0.3 M), whereas equimolar concentrations of glycerol increased primarily the D, (quasithreshold dose) with little increase in D0. The distribution of Chinese hamster ovary cells within the cell cycle was unaffected by the presence of 0.3 M glycerol in the culture medium. However, the polyols erythritol and sorbito! caused a small but significant loss of cells from the heat-resistant d compartment. The cell cycle redistribution with prolonged incubation (6 hr) in polyol-supplemented medium is expected to increase the heat sensitivity of the perturbed cell population; the observed heat protection by polyols suggests that heat resistance in the presence of polyols is not an artifact of an asynchronous cell system. Instead, the data identify a family of heat-protective compounds that may occur naturally in mammalian cells.

Stability of proteins in the presence of polyols estimated from their guanidinium chloride-induced transition curves at different pH values and 25 degrees C.

We have recently concluded from the heat-induced denaturation studies that polyols do not affect deltaG(D) degrees (the Gibbs free energy change (deltaG(D)) at 25 degrees C) of ribonuclease-A and lysozyme at physiological pH and temperature, and their stabilizing effect increases with decrease in pH. Since the estimation of deltaG(D) degrees of proteins from heat-induced denaturation curves req...

Protective Effect of Trehalose Against H2O2-induced Cytotoxicity and Oxidative Stress in PC-12 Cell Line and the Role of Heat Shock Protein-27

Background: Oxidative stress has been shown to be an important factor, which plays a significant role in the pathogenesis of neurodegenerative disorders. Heat Shock Protein-27 (HSP-27) has been implicated in antioxidant responses against oxidative stress. Trehalose is a natural disaccharide widely used in a variety of food products with demonstrated protective effects against several neurodegen...

Effect of Soluble HLA Class I Molecule on NK/LAK Cells Activation Induced by Poly I:C

Background:  Natural Killer cells express killer inhibitory receptors specific for HLA-class I molecules.  These receptors could induce signals that determine NK cells ability to mediate cytotoxicity.  Purified soluble form of HLA class I molecules (sHLA) could bind to NK cell receptors and down-regulate the NK killer function. Objective:  To evaluate the influence of sHLA and two monoclonal an...

Hyperthermic Enhancement of cis-Diammine-1,1-cyclobutane Dicarboxylate Platinum(II) Cytotoxicity in Human Leukemia Cells in Vitro1

Hyperthermic enhancement of cis-diammine-l,l-cyclobutane dicarboxylate platinum(II) (carboplatin) cytotoxicity was studied in vitro in .IM, a human acute lyntphoblastic leukemia cell line. Corrected for direct heat toxicity, hyperthermia enhanced carboplatin killing at the clinically relevant temperatures of 40.5°and 41.8°C.The thermal enhancement ratios at these temperatures were 1.89 and 33...
